    Editorial: Molecular mechanisms of flowering plant reproduction

    Plant reproduction is an intricate process important for the survival of all dominant autotrophs and critical in agriculture and stable food production as the basis of our diet. Angiosperms undergo extreme transformations during their ontogeny, leading to the reproductive transition. During this process, several steps are needed, including transforming a shoot apical or a lateral vegetative meristem into an inflorescence meristem with flowering competence. Floral meristems are formed from the latter accompanied by cell differentiation leading to gamete formation in specialized reproductive floral organs. Gamete interaction relies on successful pollination, whether that occurs via biotic or abiotic vectors. In addition, fertilization requires numerous molecular and hormonal signals in place and results in proper pollen tube growth, zygote viability, and seed formation. This Research Topic addresses some of the most outstanding discoveries on angiosperm reproduction in model species like Arabidopsis thaliana and Oryza sativa and crops like cauliflower, cassava, citrus, and sugarcane, among others.     Thienyl- and bithienyl-1,3-benzothiazoles 1 and 2 were synthesised by reacting various formyl thienyl and bithienyl derivatives with o-aminobenzenethiol in moderate to excellent yields. Evaluation of the solvatochromic and fluorescence properties of these compounds was carried out.     Platelet lysate cell-laden hydrogel-coated suture threads for tendon repair

    Tendon injuries constitute a major healthcare burden owing to the limited healing ability of these tissues and the poor clinical outcomes of surgical repair treatments. Recent advances in tendon tissue engineering (TTE) strategies, particularly through the use of biotextile technologies, hold great promise toward the generation of artificial living tendon constructs. We have previously developed a braided construct based on suture threads coated with gelMA:alginate hydrogel encapsulating human tendon cells. These cell-laden composite fibers enabled the replication of cell and tissue-level properties simultaneously. Based on this concept, in this study we explored the use of platelet lysate (PL), a pool of supra-physiological concentrations of growth factors (GFs), to generate a hydrogel layer, which is envisioned to act as a depot of therapeutic factors to induce tenogenic differentiation of encapsulated human adipose stem cells (hASCs). For this purpose, commercially available suture threads were first embedded in a thrombin solution and then incubated in PL containing hASCs. Herein, thrombin induces the gelation of PL and consequent hydrogel formation. After coating suture threads with the mixture of PL-ASCs, cells were found to be viable and homogeneously distributed along the fibers. Strikingly, hASCs encapsulated within the PL hydrogel layer around the suture thread were able to sense chemotactic factors present in PL and to establish connections between adjacent independent fibers, suggesting a tremendous potential of PL cell-laden hydrogel fibers as building blocks in the development of living constructs aimed at tendon repair applications.info:eu-repo/semantics/publishedVersio

    (Oligo)thienyl-imidazo-benzocrown ether derivatives : synthesis, photophysical studies and evaluation of their chemosensory properties

    A series of novel (oligo)thienyl-imidazobenzocrown ethers were synthesised through a simple method and evaluated as fluorimetric chemosensors for transition metal cations. Interaction with Ni2+, Pd2+, and Hg2+ in ACN/DMSO solution (99:1) was studied by absorption and emission spectroscopy. Chemoselectivity studies in the presence of Na+ were also carried out and a fluorescence enhancement upon chelation (CHEF) effect was observed following Hg2+ complexation.     Heterocyclic Aldehydes Based on Thieno[3,2-b]thiophene Core: Synthesis and Preliminary Studies as Ion Optical Chemosensors

    Heterocyclic aldehydes show a variety of optical properties and the versatility of their reactivity allows them to yield a wide range of more complex compounds, with application in areas such as medicinal, materials and supramolecular chemistry. The biological and environmental relevance of certain molecules and ions turns them into targets for the design of molecular recognition systems. Recently, heterocyclic aldehydes have been reported in the literature as ion chemosensors. Following the group's work on optical chemosensors, for the detection and quantification of ions and molecules with environmental and medicinal relevance, this work reports the synthesis and characterization of two heterocyclic aldehydes based on the thieno[3,2-b]thiophene core, by Suzuki coupling, as well as the synthesis of the corresponding precursors. Preliminary chemosensory studies for the synthesized heterocyclic aldehydes in the presence of selected cations were also performed, in solution, in order to determine their potential application as optical chemosensors

    Antibiotic free selection for the high level biosynthesis of a silk-elastin-like protein

    Silk-elastin-like proteins (SELPs) are a family of genetically engineered recombinant protein polymers exhibiting mechanical and biological properties suited for a wide range of applications in the biomedicine and materials fields. They are being explored as the next generation of biomaterials but low productivities and use of antibiotics during production undermine their economic viability and safety. We have developed an industrially relevant, scalable, fed-batch process for the high level production of a novel SELP in E. coli in which the commonly used antibiotic selection marker of the expression vector is exchanged for a post segregational suicide system, the separate-component-stabilisation system (SCS). SCS significantly augments SELP productivity but also enhances the product safety profile and reduces process costs by eliminating the use of antibiotics. Plasmid content increased following induction but no significant differences in plasmid levels were discerned when using SCS or the antibiotic selection markers under the controlled fed-batch conditions employed. It is suggested that the absence of competing plasmid-free cells improves host cell viability and enables increased productivity with SCS.     Influence of seasonality on the quality of oysters from the Sado and Mira rivers

    There is a lack of knowledge about the influence of seasonality on the microbial and physicochemical quality of oysters in Sado and Mira rivers. Water, sediment, and oysters (Crassostrea angulata and Crassostrea gigas) were collected for microbiological, nutritional, and sensory analyses. The microbiological water quality and the oyster shell contamination were better during the warmer months. No seasonal effect was observed on sediments and on oyster meat. A good physicochemical and nutritional quality was also observed, with high content of polyunsaturated fatty acids, including omega-3 fatty acids, resulting in good lipid quality indices. From the sensory evaluation, both oysters' species were well scored and presented the highest scores (4) in parameters such as cream-ivory colour, sea smell, firmness and juiciness. These attributes denote the freshness degree at the time of the tasting, reflecting the quality of the bivalve

    How much syntax is there in Metalinguistic Negation?

    This paper explores the syntax of unambiguous metalinguistic negation (MN) markers in European Portuguese (EP) with the main goal of demonstrating the syntactic import of MN. Taking the EP facts as a means to gain insight into the grammatical encoding of MN in natural language, the paper shows that unambiguous MN markers split into two types: peripheral and internal. This split is confirmed by their contrasting behavior with respect to different syntactic tests, e.g.: availability in isolation and nominal fragments; ability to take scope over negation and emphatic/contrastive high constituents; compatibility with VP Ellipsis. Peripheral MN markers respond positively to all the tests, whereas internal ones respond negatively. These facts are derived from a syntactic analysis where CP plays a central and unifying role. It is proposed that while the cross-linguistically pervasive peripheral MN markers directly merge into Spec,CP, the more unusual sentence-internal MN markers are rooted in the TP domain and reach Spec,CP by movement. The centrality of the CP field is motivated by elaborating on Farkas and Bruce’s (2010) model of polarity features. Under the hypothesis that besides the relative polarity features [same] and [reverse], there is a feature [objection] that singles out MN declaratives among responding assertions, this is taken to be the edge feature that drives unambiguous MN markers into the CP space.info:eu-repo/semantics/publishedVersio

    Production of xylooligosaccharides from Brazilian Syrah grape pomace flour

    BACKGROUND: The aim of this work was to determine the most favorable conditions for the production of xylooligosaccharides (XOS) from Brazilian Syrah grape pomace. Chemical processes were performed using a rotatable central composite design, where the concentration of sulfuric acid or concentration of sodium hydroxide and grape pomace flour: solvent mass ratio were the dependent variables. Enzymatic production was also evaluated using xylanase produced by Aspergillus niger 3T5B8 and Viscozyme® enzymatic commercial cocktail. RESULTS: Chemical extraction allowed to recover 21.8 to 74.6% and 5.2 to 96.3% of total XOS for acid and alkaline processes, respectively. Enzymatic production using xylanase extracted up to 88.68 ± 0.12% of total XOS and up to 84.09 ± 2.40% with Viscozyme®. CONCLUSION: The present study demonstrated different feasible methods to produce high added value molecules, the xylooligosaccharides, from Syrah grape pomace flour, valorizing this major by‐product. The limitations identified include insufficient metadata about registered learning objects and lack of support for intelligent applications.
